Myocardial Triacylglycerol Metabolism: The Role of ‘Competition’ Between Different Substrates

چکیده

Triacylglycerols have the potential to supply significant fatty acids for ATP production by the myocardium, yet at present the exact contribution of triglyceridederived fatty acid to the energy economy of the myocardium is unresolved. Chylomicrons and very-low-density lipoproteins both add to energy production but the degree of provision may be related to particle size of individual lipoproteins and is less affected by changes in the prevailing concentration of triacylglycerols than non-esterified fatty acids. The myocardium may represent a significant ‘sink’ for both plasma fatty acids and triacylglycerols as the heart possesses a high capillary density and high capillary surface area and may therefore represent a tissue for which ‘spillover’ of the products of lipolysis may make a limited donation to plasma non-esterified fatty acid, and triacylglycerol-derived fatty acids are partitioned towards β-oxidation to a greater extent than tissue-esterified lipids.

Regulation of myocardial triacylglycerol synthesis and metabolism.

Studies showing a correlation of excess myocardial triacylglycerol stores with apoptosis, fibrosis, and contractile dysfunction indicate that dysregulation of triacylglycerol metabolism may contribute to cardiac disease. This review covers the regulation of heart triacylglycerol accumulation at the critical control points of fatty acid uptake, enzymes of triacylglycerol synthesis, lipolysis, an...

Microsomal and lysosomal enzymes of triacylglycerol metabolism in rat placenta.

The placenta plays a major role in transporting lipid to the developing foetus. Since previous studies have suggested that placental lipid transport involves intermediate esterification steps, we investigated selected microsomal and lysosomal enzymes of triacylglycerol metabolism in rat placenta. Between gestational days 10 and 14, microsomal phosphatidic acid phosphatase specific activity was ...

Kidney Cortex Tubules

Triacylglycerol metabolism has been studied in kidney cortex tubules from starved rats, prepared by collagenase treatment. Triacylglycerol was determined by a newly developed fully enzymic method. Incubation of tubules in the absence of fatty acids led to a decrease of endogenous triacylglycerol by about 50% in 1 h. Addition of albuminbound oleate or palmitate resulted in a steady increase of t...

Triacylglycerol infusion improves exercise endurance in patients with mitochondrial myopathy due to complex I deficiency.

BACKGROUND A high-fat diet has been recommended for the treatment of patients with mitochondrial myopathy due to complex I (NADH dehydrogenase) deficiency (CID). OBJECTIVE This study evaluated the effects of intravenous infusion of isoenergetic amounts of triacylglycerol or glucose on substrate oxidation, glycolytic carbohydrate metabolism, and exercise endurance time and energy state of musc...

Hormone-Induced Variation of Phosphatidate Phosphohydrolase Activity in Various Tissues of Pregnant Rats

Variations in phosphatidate phosphohydrolase (PAP) activity and triacylglycerol concentration were measured in pregnant and hormone-treated non-pregnant female rats. PAP activity in adipose tissue was elevated by 61% during pregnancy. The increase in the enzyme activity was paralleled with a rise in serum triacylglycerol concentration (44%). Estradiol injecting into non-pregnant rats increased ...
